Tissue Banking - DNA preservation

 

A. Genomic DNA isolated from formalin-fixed paraffin-embedded (FFPE) rat tissue biopsies (1 mm diameter by 3 mm). Each sample lane represents 1/25th total recovered tissue DNA. Lane 1, molecular-weight marker(1 kb plus ladder, LifeTechnologies);lanes 2-6 are brain, kidney, liver, pancreas, and spleen (respectively); lane 7 is lambda DNA digested with Hind III. The prevalent size of genomic DNA visualized with ethidium bromide is in excess of 9-12 kilobase pairs (i.e., well preserved).

B. PCR amplification of a segment of glyceraldehyde-6-phosphate dehydrogenase (GAPDH) from genomic DNAs isolated from FFPE rat tissue biopsies. Lane 1, molecular-weight marker(1 kb plus ladder, LifeTechnologies); lane 2, water control (no DNA in reaction); lanes 3-7 are brain, kidney, liver, pancreas, and spleen (respectively). Each lane represents 1/5th of each PCR reaction using pan-specific GAPDH primers targeted to a region conserved from avian species (chicken and Pekin duck) to human.
